ABSTRACT Nanoscale cerium dioxide (CeO2, nanoceria) possesses notable redox activity, which is actively used in advanced biomedical applications. The low toxicity, high biocompatibility and antioxidant activity of nanoceria make it a new generation nanozyme with a unique activity. Combination of nanoceria with various biologically active substances results in organic-inorganic nanocomposites possessing enhanced activity. Here, we synthesized a novel organic-inorganic hybrid material (Mil-CeO2) based on 2-(2-carboxylatoethyl)-1,1,1-trimethylhydrazinium and nanoceria, which has an ultra-small particle size, high antioxidant activity and pronounced biological activity. The analysis of cytotoxicity of the composite did not reveal any negative effects on the NCTC L929 mouse fibroblasts at concentrations below 10 mM. It was shown that the nanocomposite did not cause morphological changes in cells, or lead to cell death and mitochondrial membrane potential disruption, while maintaining viability in mouse fibroblasts in vitro. Additionally, we showed that Mil-CeO2 is capable of protecting cells from hydrogen peroxide (H2O2)-induced or radiation-induced oxidative stress.

Синтез и исследование биосовместимости нанокомпозита оксид церия-милдронат in vitro

АННОТАЦИЯ Наноразмерный диоксид церия (CeO2, нанооксид церия) обладает заметной окислительно-восстановительной активностью, которая активно используется в передовых биомедицинских приложениях. Низкая токсичность, высокая биосовместимость и антиоксидантная активность наноцерия делают его нанозимом нового поколения с уникальной активностью. Сочетание нанооксид церия с различными биологически активными веществами приводит к получению органо-неорганических нанокомпозитов, обладающих повышенной активностью. Здесь мы синтезировали новый органо-неорганический гибридный материал (Mil-CeO2) на основе 2-(2-карбоксилатоэтил)-1,1,1-триметилгидразиния и нанооксид церия, обладающий сверхмалым размером частиц, высокой антиоксидантной активностью и выраженной биологической активностью. Анализ цитотоксичности композита не выявил отрицательного действия на фибробласты мыши линии NCTC L929 при концентрациях ниже 10 мМ. Показано, что нанокомпозит не вызывает морфологических изменений в клетках, не приводит к гибели клеток и нарушению потенциала митохондриальной мембраны, сохраняя при этом жизнеспособность фибробластов мыши in vitro. Кроме того, мы показали, что Mil-CeO2 способен защищать клетки от окислительного стресса, вызванного перекисью водорода (H2O2) или облучением.
